To optimize a protocol for the extraction and an in-depth analysis of the soluble protein fraction of two nonaxenic toxin-producing cyanobacteria Cylindrospermopsis raciborskii (hepatotoxin-producing), and Raphidiopsis sp. (neurotoxin-producing), using two-dimensional polyacrylamide gel electrophoresis (2D-PAGE).

Methods and Results:

The soluble protein fractions from strains of C. raciborskii and Raphidiosis sp. with different toxicity phenotypes were analysed by 2D-PAGE. Specific protocols were optimized specifically for

each strain. Between 500 and 700 sharp protein spots were distinguished in a single 4-7 pH range 2D-PAGE for each cyanobacterium. Comparison of the protein ID-8 maps of C. raciborskii CS-505 (a cylindrospermopsin-producing Sotrastaurin order strain) and Raphidiopsis sp. D9 (saxitoxin-producing strain) against the nontoxic C. raciborskii strain CS-509 revealed many unique proteins in each protein map. We confirmed that the resolved

“OBJECTIVE: Rosai-Dorfman disease is a rare benign histiocytic disease of unknown origin that arises predominantly in lymph nodes with generalized fever and malaise but can affect a variety of organs. We describe a case of isolated Rosai-Dorfman disease causing thoracic cord compression.

CLINICAL PRESENTATION: A 24-year-old man presented with progressive spastic paraparesis. A magnetic resonance scan revealed an anteriorly placed extradural lesion of the T4-T7 thoracic spine causing cord compression. He was systemically well with no other disease.

INTERVENTION: The patient made a complete recovery after a limited T4-T7 laminectomy and biopsy of the lesion. Repeat magnetic resonance scan at 6 months revealed a further posteriorly placed lesion at the T8/9 level. More extensive posterior surgery was carried out with subtotal resection of the lesion with pedicle screw fixation.

“In 2007, the U. S. National Academy of Sciences, National Research Council (NRC), issued the report Toxicity

Testing in the 21st Century: A Vision and a Strategy. This report, which was commissioned by the U. S. Environmental Protection Agency (EPA), called for the U. S. EPA to develop a new approach for how this agency evaluates the toxicity of compounds. The report recommended that the agency move from its current testing system, which is based largely on traditional toxicology, to a testing system that is based primarily on human cell lines and in vitro systems. Successful implementation of the report’s vision and strategy will require that scientists, lawyers, and policymakers work together to bridge the gap among disciplines. An important step in building this bridge requires an analysis of the U. S. legal system that frames toxicity testing. If the U. S. laws, regulations, and policies erect barriers that would prevent or impede the U. S. EPA adoption of the NRC vision and strategy, it is important to identify these challenges.

RESULTS: Median deinnervation interval was 6 months. Seventeen patients improved from grade 0 to grade 3 or more. There was no significant difference between the coordinates for maximum activity, when the cortical activity following elbow flexion of the patients who improved was compared with the control arm activity. There was reduction in FA and ADC values in patients who have improved following surgery when compared to controls. We present a patient with an EVAR infection manifesting with juxtarenal aortic rupture as a result of a urinary tract infection. “A 2-D DIGE approach allowed the characterization of the intramacrophagic proteome of the intracellular pathogen Brucella suis at the late stage of in vitro infection by efficient discrimination between bacterial and host cell proteins. Using a subtraction model, a total of 168 proteins showing altered concentrations in comparison with extracellularly grown, stationary-phase bacteria were identified.
